Procedure:
The allantoic fluids of 10-11-day-old embryonated eggs (30 eggs for seed samples) are inoculated with 0.5 mL of sample. The eggs are incubated for three days at 34-36 °C and 50-70% relative humidity; they are candled at approximately 24, 48, and 72 hours after inoculation. After the 72-hour candling, 2-5 mL of allantoic fluids from each set of 3-5 eggs are pooled and then tested for hemagglutinating viruses, using appropriate positive and negative controls. The pools are passaged via the allantoic route into four eggs each. After a second 3-day incubation, allantoic fluids are pooled according to the initial inoculum, and the embryos are examined. The pools are tested at two temperature conditions (room temperature and 2-8 °C) for hemagglutinating viruses with both guinea pig and chicken red blood cells. The sample is satisfactory if all egg deaths (not attributed to inoculation trauma) are negative for adventitious agents, there is no evidence of gross abnormalities in the embryos, and the samples display no hemagglutinating activity with the chicken or guinea pig red blood cells at either temperature.
